The PCR-CRISPR Dx: Sickle Cell Detection Kit is designed to introduce students to the application of CRISPR and PCR technologies for the molecular diagnosis of sickle cell disease (SCD). This hands-on investigation guides students through the complete workflow used in modern genetic testing, from guide RNA (gRNA) design and validation to experimental analysis.

In this activity, students assume the role of clinical scientists tasked with identifying which of four simulated patient samples contains the sickle cell mutation. Using PCR- and CRISPR-based detection, and gel electrophoresis, students analyze DNA samples and interpret experimental results to determine the disease status of each case. The kit is enough for 24 students.

The kit provides opportunities to:

·         Learn the principles of CRISPR and PCR technologies, including gRNA design and target specificity

·         Validate gRNA sequences using genome databases and bioinformatics tools

·         Perform PCR and CRISPR technologies, and analyze results using gel electrophoresis
